Technosphere® Technology

Technosphere® technology is not limited to insulin delivery. It represents a versatile drug delivery platform that allows the pulmonary administration of therapeutics currently requiring administration by injection. Technosphere technology offers several competitive advantages over other pulmonary drug delivery systems. Most notably, the pharmacokinetic profiles of drugs inhaled as Technosphere formulations approximate intra-arterial administration that is characterized by very rapid systemic absorption into the arterial circulation. First pass metabolism and peripheral degradation are also by-passed. Additionally, Technosphere technology offers the ability to conduct true placebo controlled studies by inhalation of “blank” Technosphere particles.

Technosphere formulations have been prepared from a diverse assortment of drugs that represent a broad range of physicochemical characteristics. For example, drugs ranging in molecular weight from 500 to 140,000 Daltons have been successfully adsorbed onto Technosphere particles. Anionic and cationic drugs, hydrophobic and hydrophilic drugs, protein and peptide drugs, and small molecule drugs are included in this group. Systemic drug delivery using many of these formulations has been demonstrated in a rat model of pulmonary drug delivery. Clinical drug delivery has been demonstrated with PTH/ Technosphere, sCT/Technosphere, and GLP-1/Technosphere formulations.

In addition to using the current Technosphere® particle technology to deliver a variety of different drugs, the design and preparation of novel particles is also an active area of Research and Development. Thus, our particle formation and formulation expertise is being expanded to include new particles with the potential to meet an assortment of drug delivery challenges.

While the technology is focused on pulmonary drug delivery, research has also demonstrated that proteins (including insulin) can be stabilized against degradation when formulated with Technosphere particles. This property has the potential to allow the development of fragile drug candidates that would not otherwise be feasible. Together, the assets associated with Technosphere technology have the potential to provide impressive drug delivery solutions across a wide variety of therapeutic areas encompassing a number of diverse and disease states.
